Abstract   Cytokine, a messenger of the immune system, may inhibit cell growth and proliferation and enhance cytolytic activity by modulating immune responses. Extensive studies using 12 cytokines(human recombinant IPN-e, IFN-y, IL-la, IL-l¥â, IL-2, IL-3, IL-4, IL-6, GM-CSF, M-CSF TNF-a, and TNF-¥â) were performed in order to investigate the antiproliferative and cytolytic activity of the various cytokines on human neuroblastoma cell lines SK-N-SH, IMR-32, and SK-N-MC. The antiproliferative activities were assayed by H-thymidine uptake test and the cytolytic activities by 51Cr release assay. Among the 12 cytokines, IFN-a, IFN-y, and IL-2 revealed remarkable antiproliferative and cytolytic activity and these activities varied among different cell lines. The IFN-¥ã treated natural killer cells did not show any significant increase in cytolytic activity as compared to that of the untreated natural killer cells, and the same results were obtained with monocytes. This cytolytic activity was significantly increased when natural killer cells were co-cultured with supernate of IFN-¥ã-treated monocytes. The same result was obtained with IFN-a except that the IFN-a-treated natural killer cells also showed a relatively significant increase in cytolytic activity. As was previously reported, IL-2 enhanced cytolytic activity by stimulating natural killer cells. Based on the above results, it seems reasonable ta say that the IFN-¥ã, IFN-a, and IL-2 have a considerable immunologic effect on various subtypes of peripheral mononuclear cells, and the cytolytic activity of IFN-¥ã and IFN-a on human neuroblastoma cell line is mediated mostly through manocyte-natural killer cell pathway, although it is possible that IFN-a could also stimulate natural killer cell activity in a direct way.
